Submitter rationale: The FANCG c.398C>T (p.P133L) variant has been reported in heterozygosity in at least two individuals with breast or ovarian cancer (PMID: 29905759, 32546565). It has also been reported in at least one healthy control individual (PMID: 32546565). This variant was observed in 2/34576 chromosomes in the Latino population according to the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654) and has been reported in ClinVar (Variation ID: 940932). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive.
