Uncertain significance for Hereditary spastic paraplegia 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014946.4(SPAST):c.526C>A (p.Arg176Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with serine at codon 176 of the SPAST protein (p.Arg176Ser). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and serine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with clinical features of hereditary spastic paraplegia (Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
