This variant lies in the BRIP1 gene (transcript NM_032043.3) at coding-DNA position 40, where A is replaced by G; at the protein level this means replaces lysine at residue 14 with glutamic acid — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces lysine with glutamic acid at codon 14 of the BRIP1 protein (p.Lys14Glu). The lysine residue is highly conserved and there is a small physicochemical difference between lysine and glutamic acid. This variant has not been reported in the literature in individuals with BRIP1-related disease.
