Uncertain significance for Familial Mediterranean fever — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000243.3(MEFV):c.803C>A (p.Ala268Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 268 of the MEFV protein (p.Ala268Asp). This variant is present in population databases (rs104895123, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with MEFV-related conditions. ClinVar contains an entry for this variant (Variation ID: 940448). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
