Uncertain significance for Colorectal cancer, susceptibility to, 10 — the classification assigned by St. Jude Molecular Pathology, St. Jude Children's Research Hospital to NM_002691.4(POLD1):c.745G>A (p.Asp249Asn), citing St. Jude Assertion Criteria 2020. This variant lies in the POLD1 gene (transcript NM_002691.4) at coding-DNA position 745, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 249 with asparagine — a missense variant. Submitter rationale: The POLD1 c.745G>A (p.Asp249Asn) missense change is absent in gnomAD v2.1.1 (PM2_supporting; https://gnomad.broadinstitute.org/). Five of seven in silico tools predict a benign effect of this variant on protein function (BP4), but to our knowledge these predictions have not been confirmed by functional assays. To our knowledge, this variant has not been reported in the literature in individuals with POLD1-related disease. In summary, this variant meets criteria to be classified as of uncertain significance based on the ACMG/AMP criteria: PM2_supporting, BP4.
