Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004387.4(NKX2-5):c.91G>A (p.Gly31Arg), citing Ambry Variant Classification Scheme 2023: The p.G31R variant (also known as c.91G>A), located in coding exon 1 of the NKX2-5 gene, results from a G to A substitution at nucleotide position 91. The glycine at codon 31 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
