Uncertain significance for Multiple endocrine neoplasia type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004064.5(CDKN1B):c.449G>A (p.Gly150Glu), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with glutamic acid at codon 150 of the CDKN1B protein (p.Gly150Glu). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and glutamic acid. This variant has not been reported in the literature in individuals with CDKN1B-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamic acid amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function.
