Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.3299G>A (p.Gly1100Asp), citing Ambry Variant Classification Scheme 2023: The p.G1100D variant (also known as c.3299G>A), located in coding exon 20 of the ALK gene, results from a G to A substitution at nucleotide position 3299. The glycine at codon 1100 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
