This variant lies in the LMNA gene (transcript NM_170707.4) at coding-DNA position 590, where T is replaced by C; at the protein level this means replaces leucine at residue 197 with proline — a missense variant. Submitter rationale: This sequence change replaces leucine with proline at codon 197 of the LMNA protein (p.Leu197Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individuals with LMNA-related conditions (PMID: 29237675, Invitae). In at least one individual the variant was observed to be de novo.
