This variant lies in the ACVRL1 gene (transcript NM_000020.3) at coding-DNA position 1419, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 473 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant disrupts the C-terminus of the ACVRL1 protein. Other variant(s) that disrupt this region (p.Arg479*) have been determined to be pathogenic (PMID: 15024723, 15065824, 15517393, 15712271, 16429404, 16540754, 18673552, 21158752, 23722869). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with ACVRL1-related conditions. This variant is not present in population databases (ExAC no frequency).
