Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001164508.2(NEB):c.17023C>T (p.Arg5675Cys), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the NEB gene (transcript NM_001164508.2) at coding-DNA position 17023, where C is replaced by T; at the protein level this means replaces arginine at residue 5675 with cysteine — a missense variant. Submitter rationale: Variant summary: NEB c.17023C>T (p.Arg5675Cys) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 9.4e-06 in 213292 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.17023C>T in individuals affected with Nemaline Myopathy 2 or other NEB-related disorders and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and both classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
