Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_153676.4(USH1C):c.952C>A (p.Leu318Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with isoleucine at codon 318 of the USH1C protein (p.Leu318Ile). The leucine residue is weakly conserved and there is a small physicochemical difference between leucine and isoleucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with USH1C-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
