Uncertain significance for Muscular dystrophy-dystroglycanopathy (congenital with brain and eye anomalies), type A14; Autosomal recessive limb-girdle muscular dystrophy type 2T; Muscular dystrophy-dystroglycanopathy (congenital with intellectual disability), type B14 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_021971.4(GMPPB):c.510G>C (p.Lys170Asn), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the GMPPB gene (transcript NM_021971.4) at coding-DNA position 510, where G is replaced by C; at the protein level this means replaces lysine at residue 170 with asparagine — a missense variant. Submitter rationale: This sequence change replaces lysine with asparagine at codon 170 of the GMPPB protein (p.Lys170Asn). The lysine residue is highly conserved and there is a moderate physicochemical difference between lysine and asparagine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with GMPPB-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
