NM_177438.3(DICER1):c.1694A>T (p.Asp565Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D565V variant (also known as c.1694A>T), located in coding exon 9 of the DICER1 gene, results from an A to T substitution at nucleotide position 1694. The aspartic acid at codon 565 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
