Uncertain significance for Cardiomyopathy, familial restrictive, 3; Hypertrophic cardiomyopathy 2; Dilated cardiomyopathy 1D — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001276345.2(TNNT2):c.234-2A>G, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TNNT2 gene (transcript NM_001276345.2) at the canonical splice acceptor site of the intron immediately before coding-DNA position 234, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 7 of the TNNT2 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with TNNT2-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TNNT2 cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
