NM_177438.3(DICER1):c.83T>G (p.Phe28Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F28C variant (also known as c.83T>G), located in coding exon 1 of the DICER1 gene, results from a T to G substitution at nucleotide position 83. The phenylalanine at codon 28 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
