Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.249C>G (p.His83Gln), citing Ambry Variant Classification Scheme 2023: The p.H83Q variant (also known as c.249C>G), located in coding exon 1 of the AXIN2 gene, results from a C to G substitution at nucleotide position 249. The histidine at codon 83 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
