Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004385.5(VCAN):c.4861G>A (p.Glu1621Lys), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with VCAN-related conditions. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The lysine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces glutamic acid with lysine at codon 1621 of the VCAN protein (p.Glu1621Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine.
