Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.6549A>T (p.Glu2183Asp), citing Ambry Variant Classification Scheme 2023: The p.E2183D variant (also known as c.6549A>T), located in coding exon 10 of the BRCA2 gene, results from an A to T substitution at nucleotide position 6549. The glutamic acid at codon 2183 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
