This variant lies in the RTEL1 gene (transcript NM_001283009.2) at 6 bases into the intron immediately before coding-DNA position 539, where C is replaced by A. Submitter rationale: Variant summary: RTEL1 c.611-6C>A alters a non-conserved nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: One predicts the variant has no significant impact on splicing. Whereas two predict the variant weakens the canonical 3' acceptor site and one predicts the variant abolishes the 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.6e-05 in 251166 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.611-6C>A in individuals affected with Dyskeratosis Congenita (Hoyeraal Hreidarsson Syndrome) and no experimental evidence demonstrating its impact on protein function have been reported.
