Uncertain significance for Tuberous sclerosis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000368.5(TSC1):c.3444del (p.Gly1149fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the TSC1 gene (p.Gly1149Aspfs*19). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 16 amino acids of the TSC1 protein and extend the protein by an additional 3 amino acids. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with TSC1-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated for this variant, and the functional significance of the affected amino acid(s) is currently unknown.
