Uncertain significance for Hyperkalemic periodic paralysis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000334.4(SCN4A):c.3033G>C (p.Gln1011His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with histidine at codon 1011 of the SCN4A protein (p.Gln1011His). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and histidine. This variant is present in population databases (rs369874235, ExAC 0.01%). This variant has not been reported in the literature in individuals with SCN4A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
