Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.59G>A (p.Gly20Glu), citing Ambry Variant Classification Scheme 2023: The p.G20E variant (also known as c.59G>A), located in coding exon 1 of the CBS gene, results from a G to A substitution at nucleotide position 59. The glycine at codon 20 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
