NM_024422.6(DSC2):c.2456G>A (p.Cys819Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C819Y variant (also known as c.2456G>A), located in coding exon 15 of the DSC2 gene, results from a G to A substitution at nucleotide position 2456. The cysteine at codon 819 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species, and tyrosine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
