Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251486 control chromosomes. c.407C>T has been observed in a homozygous individual affected with retinal degeneration and in a compound heterozygous individual with clinical symptoms of Leber congenital amaurosis (example: Zampaglione_2020, internal data). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
