Uncertain significance for Melanoma, cutaneous malignant, susceptibility to, 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002386.4(MC1R):c.764G>A (p.Gly255Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 255 of the MC1R protein (p.Gly255Asp). This variant is present in population databases (rs760466103, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with MC1R-related conditions. ClinVar contains an entry for this variant (Variation ID: 935677). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MC1R protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects MC1R function (PMID: 27229376).
