This variant lies in the SMARCE1 gene (transcript NM_003079.5) at coding-DNA position 547, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 183 with asparagine — a missense variant. Submitter rationale: The p.D183N variant (also known as c.547G>A), located in coding exon 7 of the SMARCE1 gene, results from a G to A substitution at nucleotide position 547. The aspartic acid at codon 183 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. This variant has been detected in multiple individuals with no reported features of Coffin-Siris syndrome(Ambry internal data). Based on the supporting evidence, the association of this alteration with an increased risk of meningiomas is unknown; however, the association of this alteration with Coffin-Siris syndrome is unlikely.
