This variant lies in the DAG1 gene (transcript NM_004393.6) at coding-DNA position 551, where C is replaced by T; at the protein level this means replaces alanine at residue 184 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine with valine at codon 184 of the DAG1 protein (p.Ala184Val). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DAG1-related conditions.
