Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006269.2(RP1):c.597C>G (p.Tyr199Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Tyr199*) in the RP1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RP1 are known to be pathogenic (PMID: 11960024, 19933189). This variant is present in population databases (no rsID available, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with autosomal recessive retinitis pigmentosa (PMID: 24265693). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 935370). For these reasons, this variant has been classified as Pathogenic.
