Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001079802.2(FKTN):c.41C>T (p.Thr14Met), citing LabCorp Variant Classification Summary - May 2015: Variant summary: FKTN c.41C>T (p.Thr14Met) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4.8e-05 in 251354 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in FKTN, allowing no conclusion about variant significance. c.41C>T has been observed in an individual affected with congenital muscle dystrophy, without strong evidence of causality (example: Masri_2022). This report does not provide unequivocal conclusions about association of the variant with Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
