The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and valine. This variant is not present in population databases (ExAC no frequency). This variant has been observed to occur as somatic mosaic in an individual affected with Muckle-Wells syndrome (PMID: 24326009) and has been observed in the germline of an individual affected with cryopyrin-associated periodic syndrome (PMID: 29047407). This variant is also known as I334V in the literature. This variant has been reported to affect NLRP3 protein function (PMID: 24326009).
