This variant lies in the INF2 gene (transcript NM_022489.4) at coding-DNA position 1577, where G is replaced by A; at the protein level this means replaces cysteine at residue 526 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces cysteine with tyrosine at codon 526 of the INF2 protein (p.Cys526Tyr). The cysteine residue is weakly conserved and there is a large physicochemical difference between cysteine and tyrosine. This variant is present in population databases (rs751942076, ExAC 0.002%). This variant has not been reported in the literature in individuals with INF2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
