Uncertain significance for Retinitis pigmentosa 71; Short-rib thoracic dysplasia 10 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015662.3(IFT172):c.3844C>A (p.Gln1282Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with lysine at codon 1282 of the IFT172 protein (p.Gln1282Lys). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with IFT172-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
