Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 254 of the BCKDHA protein (p.Ala254Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with maple syrup urine disease (PMID: 10745006, 31980395; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as A209D.
