Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001369.3(DNAH5):c.7534C>G (p.Pro2512Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with alanine at codon 2512 of the DNAH5 protein (p.Pro2512Ala). The proline residue is weakly conserved and there is a small physicochemical difference between proline and alanine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with DNAH5-related conditions.
