Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.368A>C (p.Lys123Thr), citing Ambry Variant Classification Scheme 2023: The p.K123T variant (also known as c.368A>C), located in coding exon 1 of the AXIN2 gene, results from an A to C substitution at nucleotide position 368. The lysine at codon 123 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
