Submitter rationale: Variant summary: RYR2 c.9559A>T (p.Lys3187X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay. The predominant RYR2 mutational spectrum involves missense variants in individuals with Catecholaminergic Polymorphic Ventricular Tachycardia. Therefore, the impact of loss of function variants in RYR2 on disease is not well established. At-least one truncation downstream of this position has been observed at our laboratory. The variant was absent in 235202 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.9559A>T in individuals affected with Catecholaminergic Polymorphic Ventricular Tachycardia and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
