This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 44816, where C is replaced by G; at the protein level this means replaces proline at residue 14939 with arginine — a missense variant. Submitter rationale: Variant summary: TTN c.37112C>G (p.Pro12371Arg) results in a non-conservative amino acid change located in the I-band region of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant also alters a conserved nucleotide located as the first exonic nucleotide in the exonic-splice region (exon-intron boundary) of the TTN gene. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 223266 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.37112C>G in individuals affected with Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
