This variant lies in the SYNE1 gene (transcript NM_182961.4) at coding-DNA position 17545, where A is replaced by G; at the protein level this means replaces threonine at residue 5849 with alanine — a missense variant. Submitter rationale: Variant summary: SYNE1 c.17332A>G (p.Thr5778Ala) results in a non-conservative amino acid change located in one of the Spectrin repeat domains (IPR002017) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251180 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.17332A>G in individuals affected with Emery-Dreifuss muscular dystrophy 4, autosomal dominant and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
