NM_000059.4(BRCA2):c.7586del (p.Gly2529fs) was classified as Pathogenic for Hereditary breast and ovarian cancer syndrome by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 7586, deleting one base; at the protein level this means shifts the reading frame starting at glycine residue 2529, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Variant summary: BRCA2 c.7586delG (p.Gly2529AlafsX22) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 250996 control chromosomes. c.7586delG has been reported in the literature in at-least one individual affected with Breast Cancer enrolled in a case-control association study for variants in coding regions of 11 hereditary breast cancer genes in 7051 unselected breast cancer patients and 11,241 female controls of Japanese ancestry (Momozawa_2018). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as pathogenic.
