NM_001377299.1(NDUFS2):c.1105G>A (p.Ala369Thr) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the NDUFS2 gene (transcript NM_001377299.1) at coding-DNA position 1105, where G is replaced by A; at the protein level this means replaces alanine at residue 369 with threonine — a missense variant. Submitter rationale: Variant summary: NDUFS2 c.1105G>A (p.Ala369Thr) results in a non-conservative amino acid change located in the NADH-quinone oxidoreductase, subunit D domain (IPR001135) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 250706 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1105G>A in individuals affected with Mitochondrial complex I deficiency and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
