Submitter rationale: Variant summary: ALMS1 c.2196T>A (p.Ser732Arg, also known as c.2202T>A ) results in a non-conservative amino acid change located in the Alstrom syndrome repeat domain (IPR040972) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 276512 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2196T>A in individuals affected with Cardiomyopathy/Alstrom syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
