This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 349, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 117 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Variant summary: FBN1 c.349C>T (p.Gln117X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 250556 control chromosomes. c.349C>T has been reported in the literature in individuals affected with Marfan Syndrome both in the literature (Becerra-Munoz_2018) as well as in patients undergoing evaluation for Familial Aortopathy and Marfan syndrome at our reference laboratory. These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
