This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 8167, where C is replaced by T; at the protein level this means replaces proline at residue 2723 with serine — a missense variant. Submitter rationale: Variant summary: COL7A1 c.8167C>T (p.Pro2723Ser) results in a non-conservative amino acid change located in the Collagen triple helix repeat domain (IPR008160) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 249982 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.8167C>T in individuals affected with Dystrophic epidermolysis bullosa and no experimental evidence demonstrating its impact on protein function have been reported.
