Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000314.8(PTEN):c.635-8C>A, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the PTEN gene (transcript NM_000314.8) at 8 bases into the intron immediately before coding-DNA position 635, where C is replaced by A. Submitter rationale: Variant summary: PTEN c.635-8C>A alters a nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: One predict the variant abolishes a 3 acceptor site. Three predict the variant weakens a 3 acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.2e-05 in 251422 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.635-8C>A in individuals affected with Cowden Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 928615). Based on the evidence outlined above, the variant was classified as uncertain significance.
