Likely pathogenic for Hereditary breast and ovarian cancer syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_007294.4(BRCA1):c.4185+1G>C, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BRCA1 gene (transcript NM_007294.4) at the canonical splice donor site of the intron immediately after coding-DNA position 4185, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: BRCA1 c.4185+1G>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Four computational tools predict this variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 237624 control chromosomes (gnomAD). To our knowledge, no occurrence of c.4185+1G>C in individuals affected with Hereditary Breast and Ovarian Cancer and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
