Pathogenic for Mucopolysaccharidosis, MPS-IV-B — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000404.4(GLB1):c.367G>A (p.Gly123Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: GLB1 c.367G>A (p.Gly123Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 249506 control chromosomes. c.367G>A has been reported in the literature in individuals affected with Mucopolysaccharidosis Type IVB (Morquio Syndrome B) (examples: Caciotti_2007, Yoshida_1991, Sohn_2012, Lee_2014). These data indicate that the variant is likely to be associated with disease. In vitro functional studies reduced activity for the variant (Yoshida_1991).
