Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_002474.3(MYH11):c.3963+2T>C, citing ACMG Guidelines, 2015. This variant lies in the MYH11 gene (transcript NM_002474.3) at the canonical splice donor site of the intron immediately after coding-DNA position 3963, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant alters intron 30 canonical splice donor site of the MYH11 gene. Computational splicing tools suggest that this variant may disrupt RNA splicing. To our knowledge, functional studies have not been performed to investigate this prediction. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Clinical significance of loss-of-function truncation and splice variants in the MYH11 gene is not clearly established. Available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
