NM_000051.4(ATM):c.8620C>T (p.Gln2874Ter) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 8620, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 2874 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.Q2874* pathogenic mutation (also known as c.8620C>T), located in coding exon 58 of the ATM gene, results from a C to T substitution at nucleotide position 8620. This changes the amino acid from a glutamine to a stop codon within coding exon 58. This alteration was identified in an individual diagnosed with ataxia-telangiectasia (Mitui M et al. Hum Mutat, 2003 Jul;22:43-50). This variant was also identified in an individual diagnosed with breast cancer (Xie Y et al. Clin Genet, 2018 Jan;93:41-51). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
