NM_000051.4(ATM):c.5820A>C (p.Glu1940Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1940D variant (also known as c.5820A>C), located in coding exon 38 of the ATM gene, results from an A to C substitution at nucleotide position 5820. The glutamic acid at codon 1940 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
